1. This medium is selective for fused ( hybridoma ) cells. 2. The hybridomas can be grown indefinitely in a suitable cell culture medium. 3. The plasma cells in the culture eventually die from senesence, leaving pure hybridoma cells. 4. Hybridoma technology has been replaced by recombinant DNA technology, transgenic mice and phage display.5. The medium must be enriched during " in vitro " selection to further favour hybridoma growth. 6. These CD4 + CD25 + hybridomas resemble in function the naturally occurring CD4 + CD25 + regulatory cells. 7. Hybridomas can be expanded to produce large quantities of immunoglobulins with a given unique specificity ( monoclonal antibodies ).8. One problem is to single out the hybridomas from the large excess of unfused cells after the cell fusion. 9. This maintains the well-being of the hybridomas and provides enough cells for cryopreservation and supernatant for subsequent investigations. 10. IL-6 is also essential for hybridoma growth and is found in many supplemental cloning media such as briclone.
